Topical application faces a fundamental challenge: the stratum corneum blocks molecules larger than 500 Daltons from passive diffusion, and GHK-Cu has a molecular weight of approximately 340 Daltons as a free peptide but closer to 404 Daltons when copper-complexed. It can penetrate, but the percentage that reaches target depth is low. Typically 5–15% of applied concentration.
  • Microneedling with GHK-Cu serum applied immediately post-treatment bypasses the stratum corneum barrier entirely. Studies using fluorescent-tagged peptides show that microneedling at 0.5–1.5mm depth delivers 60–80% of applied peptide to the papillary and upper reticular dermis within 10 minutes of application. The microchannels created by needling close within 24–48 hours, but the peptide is already internalized by fibroblasts during that window. Injection protocols typically use 0.5–2mg GHK-Cu per treatment session, administered every 2–4 weeks for 3–6 sessions. The peptide is diluted in sterile saline or bacteriostatic water immediately before injection to prevent degradation. Shelf life post-reconstitution is 72 hours refrigerated at 2–8°C. Beyond that, copper dissociates from the peptide and oxidative degradation reduces potency.
