- GHK-Cu is one of the few peptides with documented efficacy via both topical and injectable routes, but the mechanisms and outcomes differ meaningfully. Topical application at 1–3% concentration penetrates the stratum corneum and accumulates in the epidermis and upper dermis, where it stimulates fibroblast proliferation and collagen deposition locally. Dermatological trials using 3% GHK-Cu cream applied twice daily showed measurable increases in dermal collagen density (via ultrasound imaging) after 8 weeks and visual reduction in photoaging markers after 12 weeks. Wrinkle depth decreased by 50%, skin thickness increased by 18%, and elasticity improved by 70% compared to vehicle controls.
